I'll have to ask my friend Drew about it [he wrote for Firefly and Buffy, among other cool things], and get his thoughts. There was something just the tiniest bit "off" about the cast--like they were somehow out of their element being on the big screen. Does that make sense? How could an ensemble be so perfect for TV, and not quite "hit" on the silver screen?
The good news is that the dissonant moments were very sparse, and almost always the action and the characters drew me right back in.
prettypammie pointed out one thing that bothered her about the plotting--one of those central things to the story that has to happen, but that just doesn't make sense given everything else we know about a character's motivations and behavior. No, I'm not going to tell you what it is; I'm not sure I see it the same way, though. She's probably right, though; she usually is. Infuriating, isn't it?

I didn't sense any dissonance, other than what was scripted. I mean, the crew was at odds a bit in this movie.
As for character motivations and behavior being off... If you're talking about Mal, I think I've already argued against this on the SerenityMovie.net forums
. I think his character was spot on, despite the grumblings of some.
On the other hand, if you're talking about Simon, I would agree he seemed a bit different than the TV series character of the same name.

I was a bit surprised
at Mal -- he had stated in an episode that River and her brother were a part of the crew, but in the movie, says that they are not. That doesn't jive to me.

He strikes me as internally inconsistent enough that, at the point where we supposedly were--six months after the last episode aired--Simon was getting annoying enough, and River spooky enough that she changed her mind.
D'y'know, I thought so regarding Wash's character? I may be alone on this.
Also, I was not exactly thrilled by the (plot-related) outcome of said character.
